LGD-2226 is the ancestor that got shelved. Ligand Pharmaceuticals tested it in rats, saw solid bone and muscle data, and moved on. No human trial was ever run, no human dose was ever established. If the family name caught your eye, ligandrol reached human trials and ostarine is the milder entry point. LGD-2226 is a footnote.

Would I take it?Not when the series moved on without it.

What it is

The first SARM out of Ligand Pharmaceuticals’ pipeline, before the company moved on to ligandrol and LGD-3303. The preclinical story is genuinely interesting: bone, muscle, prostate-sparing selectivity, and even male sexual-behavior assays in rats. The company moved on, and the published record does not say why. When you hear “LGD” in the gym, nobody means this one.

What the research shows

No human trials. Zero. Everything below is animal or in-vitro work. Read it as “interesting,” not “evidence it works in you.”

Solid animal work, zero human data. The published record is entirely preclinical:

LGD-2226 produced anabolic activity in muscle and bone with less effect on prostate growth in rodent models.Endocrinology 2007 · cell + rodent
In orchiectomized rats, oral LGD-2226 prevented bone loss and maintained bone quality by stimulating bone formation while inhibiting turnover.J Musculoskelet Neuronal Interact 2002 · rat · 4 months
No human clinical trial of LGD-2226 exists. No human dose or dosing schedule has been established.as of Aug 2026

Realistic protocols

No human dose has ever been tested. You will not find a real community protocol either, because almost nobody runs LGD-2226. If you have sourced something labeled LGD-2226, start by questioning whether it actually is: the odds of a grey-market capsule containing this specific shelved precursor, rather than mislabeled ligandrol or something else entirely, are not great. A third-party COA is the minimum. If the COA checks out and you still want to run it, go low, go short, pull bloodwork before and after, and plan for PCT because suppression is the class rule. Every SARM is WADA-prohibited in tested sport.

Side effects

No human side-effect data. The SARM class profile and common sense apply:

anecdotalTestosterone suppression, expected from any compound with this mechanism.
unknownZero human safety data exists. No liver, lipid, or cardiovascular profile in people. You are extrapolating from a rat.
