Skip it. There’s no human data on dihexa, none, and the two rodent papers the whole reputation was built on are compromised: the mechanism paper got retracted, the paper that introduced it carries a notice of concern. No trials, no safety profile, no human pharmacology. I’ll walk you through what actually exists, but understand that running this is signing up as the experiment.

Would I take it?No, receipts too thin.

What it is

A small angiotensin-IV-derived molecule out of Washington State University, built to drive synaptogenesis, the growth of fresh connections between neurons. You’ve seen the headline: roughly ten million times more potent than BDNF. That line is vendor copy, not a finding, and in a second I’ll show you it traces to no actual study. What real work exists was done in dishes and rodents. On the nootropic shelf it sits near semax and noopept, except those have human dosing behind them and dihexa has none.

What the research shows

No human trials. Zero. Everything below is animal or in-vitro work. Read it as “interesting,” not “evidence it works in you.”

Here’s the entire human evidence base: nothing. And the rodent base underneath it is cracked.

The 2014 paper establishing that dihexa works through the HGF/c-Met growth-factor system was retracted by the journal in 2025. The mechanism story, pulled.JPET 2025 · retraction notice
The 2013 paper that introduced dihexa and its rat cognition data carries a JPET Notice of Concern. Both pillars of the legend are flagged.JPET 2021 · notice of concern
No human trial, case series, or safety study exists. A clinicaltrials.gov search for dihexa and for PNB-0408 returns zero.as of Aug 2026
The ten-million-times-BDNF line appears in no abstract in the dihexa literature. It travels on vendor copy, not a study.as of Aug 2026

Realistic protocols

I’m not building a protocol out of rat math and a retracted paper. For the record, forum lore runs 1 to 10 mg swallowed or rubbed on transdermally, a couple times a week, and some stacks push to 30 mg. That’s lore, not evidence: the intermittent dosing is a guess about accumulation, because nobody has measured how long it lingers in a person. If you run it anyway, go low, go short, log everything, and don’t kid yourself that a growth-factor signal with zero human safety data is a free lunch.

Side effects

Nobody has measured a single side effect in a human, so this is anecdote only:

anecdotalHeadaches and brain fog, worse at the higher forum doses.
anecdotalFlat mood or lethargy, reported by a minority.
unknownA drug that tells cells to grow, with zero human safety data, earns real caution. Growth signals cut both ways.
