The lab work is genuinely interesting. PNC-27 punches selective holes in cancer cell membranes, leaves normal cells alone, and does it across a range of tumor types in vitro and in animal models. But the gap between a dish and a person is the whole question, and PNC-27 has never entered a human trial. If you are considering this for cancer support, understand that you are running an experiment with no human safety or efficacy data behind it. Talk to your oncologist first. Standard cancer protocols have hard-endpoint trial data behind them, and PNC-27 does not.

What it is

A 32-residue peptide built from a piece of p53, the tumor-suppressor protein, fused to a cell-penetrating leader sequence. The mechanism hinges on HDM-2, a protein that cancer cells overexpress on their surfaces. PNC-27 binds that surface HDM-2, recruits more of it into ring-shaped pore structures, and the cell dies by necrosis as its contents leak out. Normal cells keep HDM-2 inside the nucleus where it belongs, so PNC-27 leaves them alone. That selectivity has held in every cell line on the published record: leukemia, colon, ovarian, cervical, and pancreatic cancer cells all die, while matched normal controls survive.

The catch is that the entire story is in vitro and in animal models. No one has tested whether the peptide survives, distributes, and selects the same way in a living person.

Realistic protocols

No human dosing data exists for PNC-27. The in-vitro IC50s tell you what kills cancer cells in a dish, not what reaches a tumor in a person. Forum protocols describe subcutaneous injection near tumor sites, but that is anecdote built on in-vitro logic, not evidence. If you run it, start at the lowest volume, keep the course short, and log every injection and response. A peptide with clean selectivity in a controlled environment may not survive, distribute, or select the same way once it enters circulation.
