The most interesting compound on this shelf that nobody has tested in a person. MOTS-c is a mitochondrial peptide with clean mouse data on energy, endurance, and metabolic aging, and zero human dosing data of any kind. I find the mechanism genuinely compelling, especially for holding energy in a deficit or around fasted cardio. But nobody has run an exogenous dosing trial, so the protocol is community lore and the safety is a guess.
What it is
A 16-amino-acid peptide encoded by your own mitochondrial DNA, one of a small family of peptides your mitochondria make and export as signals. MOTS-c activates AMPK, the cell’s energy sensor, through a folate-purine pathway, and under metabolic stress it moves into the nucleus to regulate gene expression directly. Think of it as a distress signal your mitochondria send when energy is tight, telling the rest of the cell to adapt. Endogenous levels shift with age, exercise, and metabolic health, which is how the longevity and biohacker communities found it.
The distinction matters: everything known about MOTS-c in humans is about measuring what your body already makes. Nobody has published a trial of injecting it. The mouse work is where the action results live, and those are genuinely strong for a preclinical compound: performance gains across age groups, metabolic protection, and late-life healthspan extension.

Realistic protocols
No human dose of MOTS-c has been studied, so there is no protocol to defend with data. What the community runs: 5 to 10 mg subcutaneous, two to four times per week, often timed around fasted training or during a cut, because the peptide’s own biology is exercise- and stress-responsive. Those doses are forum consensus, not a study result.
If you run it, go low, go short, and log what you feel. The case for MOTS-c is that the mechanism and the mouse data line up unusually well, not that anyone has proven an exogenous dose does anything in a person. If the energy and endurance shift does not show in the first few weeks, it probably is not coming at that dose.
Side effects
Nobody has measured a side effect of exogenous MOTS-c in a person:
