FOX04-DRI is the senolytic peptide the longevity crowd has been watching, and the animal results are genuinely interesting: old mice got younger-looking fur, better kidney function, restored fitness. But nobody has given this to a human in a published study. No dose, no pharmacokinetics, no safety profile. And one animal model flagged a real worry, worsened hemodynamics in the lungs when senescent cells were cleared. The mechanism is compelling. The evidence to act on it is not here yet.

What it is

FOX04-DRI is a designed peptide built to clear senescent cells, the damaged, non-dividing cells that accumulate with age and secrete inflammatory signals. It belongs to the senolytic class alongside dasatinib plus quercetin and fisetin, but where those work through broad kinase or flavonoid pathways, FOX04-DRI targets one specific interaction: the binding between the FOXO4 protein and p53, which is the handshake that keeps senescent cells alive. Break that handshake and the senescent cell triggers its own death by apoptosis, while healthy cells, which do not depend on that interaction, are left alone. The “DRI” stands for D-retro-inverso, a peptide engineering trick that flips the amino acids to make the molecule resistant to the enzymes that would normally chew it up in minutes.

Realistic protocols

No human dose has been validated. The community that runs FOX04-DRI typically injects 10 to 20 mg subcutaneously, once daily for 5 to 10 days, once or twice a year, treating it as a periodic senolytic reset. That dosing is extrapolated from the mouse work and forum consensus, not pharmacology. If you run it, go low, keep the course short, and log everything. The pulmonary hypertension signal from the animal work means anyone with heart or lung issues should stay away entirely. If senolytic clearance is the goal and you want a protocol with more collective mileage behind it, the dasatinib plus quercetin combination, dasatinib being a prescription leukemia drug with real toxicity of its own, is where most of the community experience sits, though that too lacks large human trials.

Side effects

No human side-effect data exists. What follows is from animal work and community reports:
