Skip it. The mechanism is genuinely interesting, an enzyme target that links fat metabolism to the NAD+ pathway, and the mouse data tells a clean story. But there is not one human trial, not one human PK measurement, not one human safety number. Vendors are selling capsules to people based entirely on what happened in fat mice. The refusal is the advice: if you want to lose weight, retatrutide and tirzepatide have massive human evidence bases and they work.

What it is

A small-molecule inhibitor of NNMT, nicotinamide N-methyltransferase, an enzyme that chews through NAD+ and S-adenosylmethionine in fat cells. Block NNMT and, in theory, you restore NAD+ levels in adipose tissue, suppress lipogenesis, and tilt fat cells toward burning instead of storing. It is a metabolic switch rather than an appetite signal, a completely different mechanism from the GLP-1 drugs that dominate this shelf.

The pitch writes itself, which is exactly the problem. Vendors jumped on the mouse data, skipped the part where someone tests it in a person, and started selling capsules. On this shelf 5-amino-1MQ sits next to compounds like retatrutide and tirzepatide that have moved tens of thousands of people through randomized trials. The gap is not small.

Realistic protocols

I am not building a protocol from mouse data and vendor capsules. Vendors sell 50 to 100 mg oral capsules, typically dosed once or twice daily, and that is where the community numbers come from. Nobody knows the human bioavailability, how long it lasts in a person, or whether the dose that worked subcutaneously in mice translates to an oral dose in a person. If you run it anyway, keep the dose low, keep the run short, and log everything. But the honest advice is to wait until someone publishes a human trial, or spend the money on a compound that has already passed that bar.

Side effects

No human side-effect data exists. The mice tolerated it, for what that is worth:
